CGRP及其表观遗传修饰在慢性疼痛中的研究进展

Research progress of calcitonin gene-related peptide and its epigenetic modifications in chronic pain

  • 摘要: 慢性疼痛作为全球致残的主要因素,其机制和治疗策略始终面临重大挑战。近年研究显示,降钙素基因相关肽(CGRP)是慢性疼痛调控的关键介质,其表达水平受到表观遗传机制的精准调控,研究CGRP表观遗传调控是寻找其参与疼痛发生发展的有效途径,这为揭示慢性疼痛的分子机制提供了全新视角。文章综述了CGRP以及其表观遗传学发现作用于慢性疼痛的机制的研究进展,包含DNA甲基化、微小RNA调节和组蛋白修饰,以期帮助临床工作者更好地理解CGRP在疼痛病理生理学中的关键作用,为疼痛治疗提供新的靶点。

     

    Abstract: Chronic pain is a major factor leading to disability worldwide, and its mechanisms and treatment strategies have always faced significant challenges. In recent years, studies have found that calcitonin gene - related peptide (CGRP) is a key mediator in the regulation of chronic pain, and its expression level is precisely regulated by epigenetic mechanisms. Investigating the epigenetic regulation mechanism of CGRP is an effective way to find out its involvement in the occurrence and development of pain, which provides a new perspective for revealing the molecular mechanism of chronic pain. This review summarizes the research progress on the mechanisms of CGRP and its epigenetic findings in chronic pain, including DNA methylation, miRNA regulation, and histone modification, helping clinicians better understand the key role of CGRP in the pathophysiology of pain and providing new targets for pain treatment.
